I'm refinishing my Fender strat in nitrocellulose, but I was wondering if it is necessary to have a primer over the sealer coat, or if its possible to just go straight on with the colour after the sealer?

It depends how nice and smooth your sealed surface is. Primer is good for putting a lot of material down that you can sand back to get a flat surface. Colour coats show up imperfections more than clear but if your prep work is ok then you're good to go.
